Archaeosomes for both cell-based delivery applications and drug-based delivery applications

Archaeosomes, or liposomes produced with one or more Archaeobacteria-specific ether lipids, are a novel kind of liposome found in Archaea. The fundamental structures of Achaean-type lipids are archaeol (diether) and/or caldarchaeol (tetraether). To make archaeosomes at any temperature in the physiological range or lower which enable thermally stable compounds to be encapsulated, traditional procedures like hydrated film sonicated, extrusion, and detergent dialysis are used. A multitude of physiological and environmental factors impact its stability. Archaeosomes are widely used as drug delivery systems for cancer vaccines, Chagas disease, proteins and peptides, gene delivery, antigen delivery, and administration of natural antioxidant compounds. The major purpose of this study was to look at how this unique carrier technology may be used in the pharmaceutical industry SOURCE : Ajay G Pise, Shilpa A Pise, 2022.  Dendritic cells, the potent antigen presenting cells, originate from the bone marrow cells and are mostly seen in the lymphoid organs of the human body. Their main purpose is to induct T-helper immune responses whenever they come across an antigenic stimulus. The presence of dendritic cells in the human cervix evoked interest in many researchers to assess their locations, distribution in cervix and to analyze their actual contribution to host immune system. In our present study, we have analyzed the actual distribution of CD1a positive dendritic cells in the benign, premalignant and malignant lesions of the human cervix. This prospective analysis was done on cervical tissue taken from hysterectomy specimens or from cervical punch biopsies. A total of 30 specimens were included in the study. The number and distribution of the dendritic cells in each of the specimens per high power field was calculated. Bleeding per vaginum was the most common presentation. Chronic non-specific cerv...

Design and development of montmorillonite-sodium alginate microbeads for oral sustained delivery of cefaclor

Drug administration via oral route is most suitable for drug delivery to patients. But this route is associated with several drawbacks such as high frequency of administration as well as gastric discomfort. To avoid these problems attempt was made to develop the sustained release system by using nanocomposite clay (MMT) and polymer (Na-Al) based microbeads for oral administration of CFR. The formulations were optimized and validated by using Box–Behnken design with 3 different levels of MMT, Na-Al and CaCl2 wherein the %EE and % drug release at 8 h were considered as dependent responses. The developed formulation was characterized by FT-IR, DSC as well as XRD. The optimized formulation was evaluated for % yield, swelling behavior, particle size, drug release and surface morphology by FESEM. The developed formulation showed physical interaction due to surface adsorption and intercalation of drug with that of MMT. The optimized formulation showed high yield, %EE and swelling behavio...

Physiotherapy considerations on Fatigue and Movements Post Modified Radical Mastectomy

Shoulder flexion, abduction, and external rotation at 90 degrees of abduction are affected after breast cancer surgery. Rehabilitation is often customised to the individual needs of every patient. Not every patient experiences the same rehabilitation, and as such, physiotherapists are able to support patients with a variety of symptoms. When selecting a type of procedure, duration, location and the type of mastectomy are essential considerations. Exercises can be recommended as early as 24 hours after surgery to maintain the shoulder range of motion and arm mobility. In restoring strength and promoting good circulation, these exercises are important. These activities can be changed to achieve new objectives as recovery progresses. In previous years, the preferred treatment for allstages of breast cancer was complete removal of the breast or radical mastectomy. Currently, various forms of mastectomy are tactically performed on the patient, based on age, menopausal status, cancer stage, ...

Conquering stress and infertility

Stress word itself causes stress for many. It is a state of mental or emotional strain. Infertility also similarly creates stress in the mind of the person suffering from it. Interestingly Stress also lead to Infertility, by altering the hormonal milieu of an individual. Treatment of infertility therefore requires interventions to reduce Stress. Diagnosing and quantifying this stress is very important. Stress reduction requires a multimodality approach. It should include pharmacological, non-pharmacological, behavioral, psychological, social, lifestyle, educational and mind-body interventions or strategies. These various approaches to reduce stress and manage infertility needs a structured clinical testing and trials. Once these interventions are routinely included in all clinics and hospitals managing infertility, their effects could be better studied. The purpose of this review is to inculcate this multimodality approach in each and every heath care provider who is involved in managi...

A comparative study based on two different stretching protocol of 30 second vs 10 second for piriformis tightness Ruchika Zade, Sakshi P. Arora, Pratik Phansopkar, Mitushi Deshmukh

Piriformis syndrome, a prevalent cause of low back discomfort around the world, is caused by muscular tightness. By serving as an external rotator, a weak hip abduction, and a weak hip flexor, the piriformis muscle promotes postural stability when walking and standing. Stretching aids in decreasing the stiffness of the Piriformis muscle at rest, as well as the risk of Sciatic compression caused by Piriformis tightness. Reduce discomfort and muscular tightness while increasing range of motion. The experiment will be a randomized trial. A total of 20 persons would be chosen for the study based on the inclusion or exclusion criteria from AVBRH & RNPC, Sawangi Meghe. The study will last six months. Twenty participants were divided into two groups. All groups received an ice pack approximately 10 minutes prior to stretching, and all groups received static stretching. Stretching is done for 30 seconds in group A and 10 seconds in group B. The key outcome measurements for the patient shou...

Identification of plant secondary metabolites as dengue envelope protein entry inhibitors through an in-silico approach

The entrance of the dengue virus to the host cell is mediated by its main envelope (E) protein. The crystal structure of E protein shows a hydrophobic pocket occupied by the detergent n-octyl-β-D-glucoside (βOG) lying in the Hinge region among domains I&II, which recognizes the hydrophobic ligand, opens and closes via a transition state in the beta-hairpin at the interface between two domains at low pH triggered conformational rearrangement necessary for fusion. We have identified fourteen plants that have been reported for anti-dengue activity. Though these plants are noted for anti-dengue activity, their molecular mechanisms are not fully explored. To know their targets, molecular docking experiments using Command Line Calculator (CLC) Drug Discovery Workbench Software have been performed against dengue virus envelope protein (DENV E) (PDB ID: 1OKE). The Glabranin and Hyperoxide have been showing docking scores (-52.89 and -51.27), nearing the standard n-Octyl-beta-D-glucopyranos...

Detecting epithelial sodium channel (ENAC) protein levels as a biomarker of hypertension

Epithelial Sodium Channel (ENaC) is a protein that plays a role in maintaining sodium levels in the body. ENaC protein is abundant in the intestines, lungs, and kidneys. ENaC protein levels were measured from specific locations where the kidneys would be a good source of sodium excretion associated with hypertension. ENaC protein is one of the biomarkers hypertension based on lifestyle because it is related to sodium intakes in the body. ENaC protein is a biomarker of hypertension because it plays a role in sodium ion transport in cells which is related to blood pressure levels. Most hypertension or high blood pressures are not diagnosed on time because they are asymptomatic and existing protocols are less effective. The method to detect ENaC protein has been developed and can be a solution to this problem. This review describes various methods to detect ENaC protein as a biomarker of hypertension.   https://jmpas.com/abstract/1037
